O'Brien was a four-year letterwinner for the Holy Cross women's basketball team, leading the squad to Patriot League titles in 1996, 1998 and 1999. A first team All-Patriot League selection in each of her final three seasons, O'Brien was named the Patriot League Player of the Year and Scholar-Athlete of the Year in both 1998 and 1999. Only the second player in school history to score over 2,000 career points, she finished her career as a Crusader with 2,075 points, 963 rebounds, 179 assists and 209 steals.

O'Brien graduated holding the top two spots on the Holy Cross single season scoring list, with 782 points as a junior and 643 points as a senior. A two-time second team Academic All-America selection, O'Brien posted 40 career double-doubles and scored 30 points in a game 14 times, including a school record 38 points against Connecticut in her junior year. O'Brien was named the Crusader of the Year in 1999, and was inducted into the New England Basketball Hall of Fame in 2003.
